I was once stopped at Hilton's car park entrance and asked by the guard to open the boot for security check. To my embarrassment, my boot was lock even after i press the "button". I had to stop my engine, took out the car and use the key to open my boot. But sometimes my boot can be opened without the key, My question is what happened or how to ensure the boot can always be opened without the keys, when the car engine is running?

The boot should not unlock without using the key. If sometimes it opens without the key it is because you have unlocked it earlier. Once you've locked the car again on the remote you will need to unlock the boot with the key again to be able to open it. It is designed to work that way, I guess to make it more secure. What happened to you was quite normal.
To answer the last part of your question, unlock the car using your remote, unlock the boot using the key, and the boot will then accessible using the button until you have locked it again.
